我在美国做基因检测（中）

上一篇文章“我在美国做基因检测（上）”中谈到了一些问题，比如基因检测到底是检测什么的？如果我要做基因检测都需要准备什么？以及基因检测的理性对待。

今天小编想谈一谈，这些基因检测公司背后的技术“大佬们”。
2017年12月2日，小编收到了23andMe公司发来的邮件，有图有真相~~~~

23andMe 告诉小编，小编的唾液样本已经到达23andMe公司的实验室。他们让小编等待6-8周才可以拿到。小编当时就疑惑了：6-8周？？？为神马这么慢！！！！
那么问题来了，基因检测真的需要这么慢吗？
23andMe检测的核心技术到底是什么？
这核心技术真的需要用这么长的时间才能检测出我的SNP（SNP介绍请看上一篇文章）基因突变吗？
一切都要从下面这张芯片说起。

就是这张不到巴掌大的小小芯片，里面固定了多达约71万个基因片段检测探针。只要加入待测者的样本，就可以通过读取反应信号来检测待测者的基因突变位点信息。这张芯片有24块样本区，也就是说，这一张芯片可以同时检测24个人的基因突变位点信息。
整个检测时间从加入样本，到信号输出所用时长只需2-3分钟。一个星期就可以检测大约2304个人的样本，当然你可以定制你个性化的芯片，可以提高每一张芯片的容量，比如可以扩充至每张芯片去检测48个、288个、甚至是1152个样本！
这种高通量的检测操作简直就是科技界的美丽奇观！

那么，这么牛X的技术是23andMe公司开发的吗？
答案是No~No~No~
23andMe公司的核心硬件技术–Genotyping，这张芯片背后的“产业大佬”是一家叫Illumina的生物公司。Illumina公司是全球领先的、专门进行开发、制造和销售用于大规模分析遗传变异和基因功能的生命科学工具及一体化系统的厂商。他们的客户遍布全球广泛的科研、政府、制药、生物技术及其他主要机构。

23andMe正是依托Illumina的定制芯片而使商业化的基因检测成为可能。23andMe在Illumina定制的芯片叫做“Infinium OmniExpress-24 Kit”，里面有23andMe公司为了自家公司的检测需求而自行设计的检测探针。也就是如果你有需要的目标基因或者基因突变位点，你是可以自行设计这张芯片中的探针的。

那么，既然这张芯片这么牛X，为什么我还要等6-8周才能拿到我的报告呢？
小编觉得有这么三点原因:
1. 你的唾液样品在加入这张芯片之前需要在样本准备室里面是需要提取DNA的。这是一个比较复杂的生物实验过程，比如生理盐水细胞冲洗啊、样本离心啊、转移到硅胶膜吸附DNA啊，还有PCR扩增等等。这都是需要实打实地一个样品一个样品的准备的。
2. 因为芯片一张可以测24个人的样本，如果这段时间邮寄到23andMe的样本不到24个怎么办？那么商业公司肯定要考虑成本的，那你的样本只能在冰箱里面保存并等待攒够了24个人再统一检测。所以说，一张芯片可以同时检测1152个样本在这个时候就很尴尬了。
3. 后期的数据分析。芯片的信号读取只是最最原始的数据。并不能够直接作为基因检测报告使用。产生的信号数据后期需要quality control和reference genome mapping等一些列pipeline操作才能够生成含有SNP的数据文件。然而你最后拿到的具有可视化的、好看的图片的报告还需要23andMe的数据科学家利用机器学习、高级算法来决定你的先祖情况以及你可能会获得某种疾病的危险概率。所以现在回头看来，小编觉得23andMe或者其它基因检测公司的未来发展好坏与否和他们是否能够召来好的数据科学家息息相关。

基因检测背后的大佬只有Illumina一个吗？
当然不是。小编查了一下美国和中国目前比较有名的基因检测公司和他们背后的硬件“大佬们”，总结了如下一张表格。

可以看出来基本上芯片供应商有两大家，Illumina和ThermoFisher。而美国基因检测偏向Illumina，国内则同ThermoFisher公司合作更多。前四个公司是现在可以检测很多基因突变位点并产生复杂性分析报告的公司。然而还有其他一些小公司只做一种或者两种的基因检测报告，比如源基因只是溯源，或者只是检测阿尔兹海默症。这种公司可能就不会需要Illumina或者ThermoFisher这些大佬们提供那么复杂的芯片，因为他们只关心DNA中若干个基因的信息即可。
